چکیده

Background Asthma is a chronic condition described by inflammation of the airways and lungs. Diesel exhaust (DE) is a major contributor to ambient particulate matter (PM) air pollution. There is rising evidence that PM acts as adjuvant on the immune responses and may lead to augmentation of allergic inflammation [1,2]. We aim to elucidate if DE increases allergen-induced inflammation and cellular immune response in the airways of atopic human subjects.

Prenatal allergen and diesel exhaust exposure and their effects on allergy in adult offspring mice

BACKGROUND Multiple studies have suggested that prenatal exposure to either allergens or air pollution may increase the risk for the development of allergic immune responses in young offspring. However, the effects of prenatal environmental exposures on adult offspring have not been well-studied. We hypothesized that combined prenatal exposure to Aspergillus fumigatus (A. fumigatus) allergen an...

Diesel exhaust augments allergen-induced lower airway inflammation in allergic individuals: a controlled human exposure study.

RATIONALE Traffic-related air pollution has been shown to augment allergy and airway disease. However, the enhancement of allergenic effects by diesel exhaust in particular is unproven in vivo in the human lung, and underlying details of this apparent synergy are poorly understood. OBJECTIVE To test the hypothesis that a 2 h inhalation of diesel exhaust augments lower airway inflammation and ...

Airway antioxidant and inflammatory responses to diesel exhaust exposure in healthy humans.

Pulmonary cells exposed to diesel exhaust (DE) particles in vitro respond in a hierarchical fashion with protective antioxidant responses predominating at low doses and inflammation and injury only occurring at higher concentrations. In the present study, the authors examined whether similar responses occurred in vivo, specifically whether antioxidants were upregulated following a low-dose DE c...

Diesel Exhaust Particles Induce Cysteine Oxidation and S-Glutathionylation in House Dust Mite Induced Murine Asthma

BACKGROUND Diesel exhaust particle (DEP) exposure enhances allergic inflammation and has been linked to the incidence of asthma. Oxidative stress on the thiol molecules cysteine (Cys) and glutathione (GSH) can promote inflammatory host responses. The effect of DEP on the thiol oxidation/reduction (redox) state in the asthmatic lung is unknown. OBJECTIVE To determine if DEP exposure alters the...

Effect of glutathione-S-transferase M1 and P1 genotypes on xenobiotic enhancement of allergic responses: randomised, placebo-controlled crossover study.

BACKGROUND Particulate pollution is associated with the occurrence of asthma and allergy. The model pollutant, diesel exhaust particles, can participate with allergens in starting and exacerbating allergic airway diseases in part by production of reactive oxygen species. Glutathione-S-transferases (GSTs) can metabolise reactive oxygen species and detoxify xenobiotics present in diesel exhaust p...
